^^Je suis pas d'accord, le diazepam a des propriétés de relaxateur musculaire tres puissant, un des plus puissant parmis les benzos que j'ai essayé .... le xanax quand a lui est tres utile pour des gens qui ont des attaques de paniques soudaines, plutot qu'une angoisse qui est la tout le temps, traitable avec du clonazepam du clorazepate ou du diazepam... le clonazepam etant eut etre plus intérréssant car il n'est pas hypnotique... donc n'endort pas comme les autres (a des doses thérapeutiques)
Sinon pour le Zolpidem, beaucoup de gens ont des hallucinations s'ils se battent contre l'envie de dormir et restent reveillés, c'est pourquoi cette molécule est assez détournée...
De plus le diazepam est peut etre le plus important thérapeutiquement car il permet de se sevrer des autres benzos, ou le sevrage de l'alcohol

This shows a few things:
*Potency has little to do with euphoria; temazepam, this third weakest benzo based on absolute receptor affinity, is one of the most prized for euphoria.
*SAR information:
- A triazolo ring around positions 1 and 2 (which makes the amide tertiary) increases potency. If the triazolo ring is methylated where it is in alprazolam and triazolam, an extreme increase in potency occurs.
- An electron attracting group at position 7 is necessary for activity. Chlorine, fluorine, and nitro groups are all suitable. Nitro groups seem to increase potency the most.
- An electron attracting group (usually halogens) at position 2' GREATLY increases potency.
*Hypnotic Dominant vs. Anxiolytic / Anticonvulsant Dominant Benzos
- If two or more benzos have the same absolute potency, the hypnotic (somewhat GABA-A subtype a1 selective) ones feel subjectively stronger.
Example: Look at the top three most absolutely potent benzos. Triazolam, alprazolam, and clonazepam all have uM values very close together, but triazolam is considered twice as potent in most mg dose charts. It packs twice the potency of xanax and kpin based mostly on perception of the effects, because it's selective for a1, a hypnotic benzo. Same with lorazepam and lormetazepam. Lorazepam actually has a higher affinity overall for GABA-A BZD receptors, but most users feel that lormetazepam packs a greater punch -- that lower doses of lormetazepam are needed to fuck you up.
- Now, the SAR data for what makes a hypnotic benzodiazepine --
a) The amide at position 1 being tertiary rather than secondary makes a benzo more hypnotic. Methyl groups (ala temazepam) and the triazolo ring (ala triazolam) both make this amide tertiary
b) Nitro as the electron attracting group at position 7 makes a benzo more hypnotic (ala nitrazepam)
c)Fluorine as the electron attracting group at position 2' makes a benzo more hypnotic (ala flunitrazepam), but a chlorine can suffice if a methylated triazolo ring is present (ala triazolam)
d)Combinations of these (ala flunitrazepam) make a benzo even more hypnotic. Sometimes only one of these features isn't enough to make a benzo hypnotic (an example would be clonazepam)."
